I don't think there's any way that SCS can check and/or temporarily set such settings, but I'll make some enquiries.

I have had this happen to me on a couple of occasions, and in differing versions of SCS (10.x), and until recently had not been able to identify what was happening.
In my case I had run a few cues in the Tech rehearsal, and then noticed that I needed to renumber them slightly, so did this (in the editor window) and returned to the playback window. When I then tried to play the next cue, it triggered, but there was no audio. I closed and reloaded the software, no luck, so I then rebooted the pc and reloaded the show, still no luck. Loading another show, all was fine, but reverting to my current show, no audio again.
Luckily the machine had a much older version of SCS (9.x) so I loaded the cue file into that, and apart from the fact that it didn't quite understand the RS232 Control Send Cues (understandable), the Audio Played perfectly and everything else was fine.
After the rehearsal, I loaded my current version of SCS again (10.6.3) and again no audio. I then noticed that the 'Master' fader below the meters was set at infinity (the lowest level/off), so I dragged it back up to its previous level, but was amazed to watch it slide itself back down to nothing! Nothing I could do would make it stay at its correct level, and checking the 'Mixer levels' Tab in the settings, the levels were correct there!
Resigned to the fact I would be using v9 to run the show, I made some changes to the cue file and ran the dress rehearsal on v9. I then tried loading the now modified cue file back into v10 and it was fine again!
In my case I would say that the problem relates to the cue file. Like other posters on here, I am using an Echo Layla card, but playing with the sync settings etc as mentioned above, had no bearing on whether audio would play, but loading differing cue files did.
I am not sure if this helps you track down this issue, but for me it was definitely the master level (audio would play back briefly whilst it was fading out) and that seemed to be triggered by something I did by renumbering the cues! It would be interested to know from others whether their 'Master' fader gets set to off when this issue occurs.

That's interesting, Graham. Regarding the Master Fader sliding by itself, I've listed below the ways in which the Master Fader can be adjusted, but most of them are independent of the cue file:
Can you reproduce this problem - eg do you still have the version of your cue file that gave you this error? If so, I can provide you with a tracing version of SCS that will help me identify what's happening with the Master Fader.
- Manually dragging the Master Fader slider
- Opening a cue file - the initial Master Fader setting is obtained from the Production Properties
- Changing the Master Fader setting in the Production Properties
- Using keyboard shortcuts that are mapped to the Master Fader (see General Options / Permanent Options / Key Mapping)
- Receiving a MIDI message assigned to adjusting the Master Fader
- Receiving a DMX message assigned to adjusting the Master Fader
Can you reproduce this problem - eg do you still have the version of your cue file that gave you this error? If so, I can provide you with a tracing version of SCS that will help me identify what's happening with the Master Fader.

Mike, when I am next at the theatre, I will see if I still have a backup of the non functional cue file - I may just possibly!
We are using MSC to fire the cues in SCS, but there should be nothing mapped to the master fader to control that - certainly I have not mapped anything, or indeed unmapped something to make it work again. Also the show was created from scratch so there should be no settings left over from a previous production. We are not using DMX control and the PC has no DMX interfacing.
